Role Overview:
The Laboratory of Photosynthesis (Ondřej Prášil’s group) at the Institute of Microbiology, Třeboň, Czech Republic, studies photosynthetic processes in cyanobacteria and other microalgae, from molecular mechanisms to biogeochemical implications. The group is currently involved in a large national project (Photomachines, 2024–2028), focusing on the development of a novel type of photosynthetic chassis for biotechnological applications. A specific task of the research group is to participate in the construction and operation of a high-throughput robotic system (AlgaScreen) for continuous cultivation, monitoring, and selection of genotypes of photosynthetic microorganisms. This system consists of various stations to measure a range of physiological parameters in detected cyanobacterial and algal colonies from liquid cultures.

Responsibilities:
The work will include designing experiments for robotic screening, optimising protocols, tuning individual sensor units, developing protocols, processing data, and post-analysis of selected mutant lines (microscopy techniques, omics, etc.).

The main aims of the project are:
- To utilize advanced ultrastructure and spectroscopy methods (e.g. microRaman) to identify various amorphous or crystalline storage compounds and other macromolecules of interest in algae and cyanobacteria
- To improve and optimize pipeline for robotic characterization of algal photophysiology using combination of biophysical methods
- To apply machine-learning in processing of large datasets from robotic and other measurements

Requirements and Qualifications:
- PhD in natural sciences/life sciences (biophysics, plant/algal/cyanobacterial physiology microbiology, molecular biology, chemistry)
- Good knowledge of standard techniques in biophysical methods (spectroscopy and spectrofluorimetric techniques in general), Raman microscopy, confocal microscopy) and in programming (Python, R) is expected
- Additional experience in genetic manipulation of algae/cyanobacteria and omics (protein mass‐spectrometry, genomics) is helpful
- Good level of English
- 1 - 4 years of research experience
